GLP Sterility Testing of Pharmaceutical Products: Methods, Validation, and Nano-Test Solutions

The sterility test is a critical microbiological analysis used to determine whether live microorganisms are present in pharmaceutical products declared to be sterile. For injectable preparations, eye drops, infusion solutions, biological products, and sterile medical devices, this test serves as the final safety barrier for patient safety. The presence of viable microorganisms in a product can lead to serious, life-threatening clinical consequences, such as sepsis.

Sterility tests conducted in accordance with Good Laboratory Practice (GLP) ensure that the results are reliable, reproducible, and acceptable to regulatory authorities. In this article, we will discuss the basic methods of pharmaceutical GLP sterility testing, the validation requirements, and Nano-Test’s accredited services in this area.

What is a sterility test, and why must it be conducted in accordance with GLP?

A sterility test is performed to verify the effectiveness of the product’s sterilization process and to demonstrate that the final product contains no viable microorganisms. However, a sterility test alone does not guarantee that a batch is sterile; this guarantee is primarily ensured through the validation of the sterilization process or the aseptic manufacturing process. The pharmacopoeias explicitly state that the test does not replace process validation but rather supports it [reference:0].

Conducting sterility tests in accordance with GLP is mandatory for the following reasons:

  • Data Reliability: GLP establishes strict requirements regarding staff training, equipment calibration, standard operating procedures (SOPs), and data integrity (ALCOA+ principles).
  • Regulatory Approval: Regulatory agencies such as the FDA, the EMA, and the TİTCK require GLP-compliant sterility test reports for marketing authorization applications.
  • Documentation of aseptic conditions: It must be demonstrated that the test was conducted under aseptic conditions and that environmental controls were monitored regularly[reference:2].

Methods for Sterility Testing

1. Membrane filtration process

Provided that the nature of the product allows for filtration, the membrane filtration method is the preferred approach [reference:3]. In this process, the product is passed through a membrane filter with a maximum pore size of 0.45 µm; microorganisms are retained on the filter, after which the filter is transferred to a suitable culture medium and incubated.

Applications: Aqueous solutions, alcohol-based or oil-based formulations, and products that are miscible with aqueous or oil-based solvents.

For antimicrobial activity: If the product exhibits antimicrobial properties, the membrane must be washed at least three times; the washing cycle may consist of no more than five wash cycles, each using 100 ml per filter.

2. Direct inoculation method

In cases where membrane filtration is not suitable (e.g., for non-filtrable, viscous, or particulate-containing products), the direct inoculation method is used. In this method, the product is transferred directly into the culture medium[reference:7]. It is suitable for products without antimicrobial activity or those in which such activity can be eliminated by dilution or inactivation.

3. Culture Media Used

Two basic culture media are used in sterility testing[reference:9]:

  • Fluid Thioglycollate Medium (FTM): It is primarily designed for anaerobic bacteria, but it also detects aerobic bacteria.
  • Soybean-Casein Digest Medium (SCDM): It is suitable for culturing both fungi and aerobic bacteria.

Incubation conditions: For FTM, 3 days at 32.5 ± 2.5 °C (in the validation test) or 14 days (in the routine test); for SCDM, 5 days at 22.5 ± 2.5 °C (validation) or 14 days (routine).

Comparison of Sterility Testing Methods

The following table summarizes the two basic sterility testing methods:

ProceedingsprincipleSuitable Product TypesUse in Antimicrobial ProductsAdvantages
Membrane FiltrationThe product is passed through a 0.45-µm filter; microorganisms are retained by the filter and transferred to the culture medium.Aqueous, alcoholic, and oily solutions; filterable products. The membrane is rinsed at least three times (max. 5 × 100 ml).Preferred method; wide range of applications.
Direct InoculationThe product is transferred directly into the culture medium.Non-filterable, viscous products containing particles. The antimicrobial effect is negated by dilution or inactivation. An alternative in cases where membrane filtration is not suitable.

Validation of Method Suitability: The Cornerstone of Sterility Testing

A sterility test is valid only if not only the correct method is selected, but it is also demonstrated that this method is suitable for the product matrix. This validation , known as a “method suitability test,” confirms that the product’s antimicrobial activity is sufficiently suppressed under the test conditions.

Validation must be performed in the following cases:

  • When a new product undergoes a sterility test,
  • If changes are made to the test conditions,
  • If the product's composition or manufacturing technology changes.

During the validation test, the antimicrobial activity of the product is evaluated by observing whether or not the test strains (maximum 100 CFU) multiply in the presence of the product. If the test strains do not multiply in the presence of the product, this indicates that the antimicrobial activity has not been inactivated, and the validation must be repeated under modified test conditions.

Important: The sterility test must be performed under aseptic conditions (laminar flow cabinet, cleanroom, or Class A isolator). The work environment must be monitored regularly to ensure compliance with GMP and GLP standards.
